John 3:5 NKJV

Jesus answered, "Most assuredly, I say to you, unless one is born of water and the Spirit, he cannot enter the kingdom of God."

Being born of water is a given for any person because it means a person is physically born (its nothing to do with baptism); the Jewish idiom is clear. But Jesus goes much much further - in keeping with the theme of the discourse of this passage -  one needs to be born of the Holy Spirit to enter the kindom of God, and this takes place when one believes in the one and only Son of God (John 3:16). Every human is born spiritually dead; thus each person needs to be regenerated (born again) to be made a live (John 6:44, 65; Romans 8:8). Regeneration requires being born of God which is where the Holy Spirit comes in. Indeed the agent of new birth is the Holy Spirit.
